Jolley learned his craft at the knee of his father Moody. The younger Jolley began walking hots for his father at age seven, later served a two year apprenticeship under him, and then took out his trainer's license in 1958. Jolley's first winner, Somnus, was bred by his father and owned by his mother. Jolley has spent most of his career on the New York and Florida circuit. He saddled his first Kentucky Derby horse, third-place finisher Ridan, in 1962. In 1980 he won the Derby with Genuine Risk, the first filly to compete in all three Triple Crown events. She placed second in the Preakness and Belmont.

Jolley's first champion, Foolish Pleasure, later became a Hall of Fame inductee. LeRoy Jolley was inducted in the Hall of Fame in 1987.
